Light Source

The light source emits a broad spectrum of light that passes through the sample. Common types include tungsten-halogen lamps for visible light, deuterium lamps for ultraviolet (UV) light, and sometimes xenon flash lamps for specialized applications. A stable and bright light source ensures consistent illumination and reduces measurement noise, which is essential for precise absorbance readings .

Monochromator or Dispersive Element

The monochromator isolates a specific wavelength of light from the source. This can be achieved using a prism, diffraction grating, or filter wheel. By selecting a narrow wavelength band, the monochromator ensures that only the desired light interacts with the sample, which is critical for accurate spectral analysis . Modern instruments often use diffraction gratings with hundreds to thousands of grooves per millimeter to disperse light efficiently .

Sample Holder

The sample holder or cuvette compartment positions the sample in the path of the selected light. Cuvettes can be made of quartz for UV measurements or plastic/glass for visible light. The holder ensures consistent path length and alignment, which is necessary for reproducible absorbance measurements .

Detector

The detector measures the intensity of light after it passes through the sample. Common detectors include photodiode arrays, photomultiplier tubes (PMTs), and charge-coupled devices (CCDs). The detector converts light energy into an electrical signal, which is then processed by the instrument's electronics or software to calculate absorbance or transmittance according to the Beer-Lambert Law .

  • Collimators (mirrors or lenses) align light rays to travel parallel through the monochromator and sample .
