A spectrophotometer consists of a light source, optical system (monochromator), sample holder, and detector, each playing a crucial role in measuring light absorption or transmittance.

Light Source

The light source provides the initial radiation for measurement. Different lamps are used depending on the wavelength range: tungsten-halogen lamps for visible light, deuterium or hydrogen lamps for ultraviolet light, and Nernst filaments or globars for infrared light. The light must be stable and intense to ensure accurate measurements .

Optical System (Monochromator)

The optical system isolates a specific wavelength from the polychromatic light. It typically includes:

  • Collimator: A lens or mirror that aligns light rays parallel to each other.
  • Entrance slit: Produces a narrow beam of light.
  • Dispersive element: A prism or diffraction grating separates light into its component wavelengths.
  • Exit slit: Selects the desired wavelength to pass through the sample. This system allows precise wavelength selection and controls the bandwidth of light reaching the sample .

Sample Holder (Cuvette)

The sample holder is where the solution is placed, usually in a cuvette. Cuvettes are made of quartz for UV measurements or glass/plastic for visible light. In double-beam spectrophotometers, a beam splitter directs light through both the sample and a reference (blank) to improve accuracy .

Detector

The detector measures the intensity of light transmitted through the sample and converts it into an electrical signal. Common detectors include photomultiplier tubes, photodiodes, and photovoltaic cells. The signal is then amplified and processed to display results as absorbance or transmittance on a digital screen or computer .

Additional Components

  • Mirrors and lenses: Direct and focus the light beam through the system.
  • Beam splitter: In double-beam instruments, divides light into sample and reference paths.
  • Signal processor: Converts the detector's electrical signal into readable data . These components work together according to the Beer-Lambert Law, allowing the spectrophotometer to determine the concentration and purity of a sample by measuring how much light is absorbed at specific wavelengths .
